Dulaglutide research

What published studies found for Dulaglutide: 8 papers, 8 in humans, with sample sizes and key results, plus 95 registered clinical trials.

A
Strong human evidence
Why: Approved drug; many large RCTs (incl. the 9,901-person REWIND trial) consistently show better glucose control and fewer cardiovascular events.

Dulaglutide (a once-weekly GLP-1 receptor agonist for type 2 diabetes) is supported by many large human RCTs showing improved blood-sugar control and, in the REWIND outcomes trial, a modest reduction in cardiovascular events; its weight-loss effect is smaller than semaglutide or tirzepatide, and gastrointestinal side effects are common.

✓ 4 positive~ 2 mixed? 2 inconclusive

Severe GI complications such as pancreatitis or bowel obstruction were similar in frequency to semaglutide and tirzepatide in one large observational study.

US compounding: FDA-approved drug. FDA-approved (Trulicity). It's a biologic-sized fusion protein; compounded versions aren't a recognised option. Status for every peptide.

Semaglutide versus dulaglutide once weekly in patients with type 2 diabetes (SUSTAIN 7): a randomised, open-label, phase 3b trial.

n = 1,201 humansOpen-label, parallel-group, phase 3b trial (SUSTAIN 7)40 weeks

In 1201 people with type 2 diabetes, dulaglutide lowered HbA1c by 1.1-1.4 points and weight by 2.3-3.0 kg at 40 weeks, both significantly less than semaglutide; GI disorders in 33-48%.

Key resultHbA1c fell 1.8 points with semaglutide 1.0 mg vs 1.4 with dulaglutide 1.5 mg (ETD -0.41; p<0.0001); bodyweight fell 6.5 kg vs 3.0 kg.
Studied
Adults with type 2 diabetes inadequately controlled on metformin monotherapy
Compared with
Dulaglutide 0.75 mg or 1.5 mg
Main outcome
Change in HbA1c and bodyweight at week 40
Sample
1201 randomized to four arms
Authors' conclusion
At low and high doses, semaglutide was superior to dulaglutide for glycaemic control and weight reduction, with a similar safety profile.
Limitations

Dulaglutide versus insulin glargine in patients with type 2 diabetes and moderate-to-severe chronic kidney disease (AWARD-7): a multicentre, open-label, randomised trial.

n = 577 humansMulticentre, open-label randomised trial (AWARD-7); dulaglutide doses masked52 weeks (primary outcome at 26 weeks)

In 577 people with type 2 diabetes and moderate-severe kidney disease, dulaglutide matched insulin glargine for HbA1c and showed slower eGFR decline and less hypoglycemia, but more nausea (up to 20%) and diarrhea (up to 17%).

Key resultAt 26 weeks LSM HbA1c change was -1.2% with dulaglutide 1.5 mg, -1.1% with 0.75 mg and -1.1% with insulin glargine; both doses were non-inferior. Symptomatic hypoglycaemia was lower with dulaglutide.
Studied
Adults with type 2 diabetes and stage 3-4 chronic kidney disease
Compared with
Daily insulin glargine, all with insulin lispro
Main outcome
HbA1c at 26 weeks (non-inferiority margin 0.4%)
Sample
577 randomized (193, 190 and 194 per arm)
Authors' conclusion
Once-weekly dulaglutide gave glycaemic control similar to insulin glargine with reduced decline in eGFR, and seems safe in moderate-to-severe CKD.
Limitations
Open-label for insulin glargine comparison.
