Doses studied in research Studied in people
What was given to people (or animals) in published studies and official labels, each linked to its source. This is not a recommendation: study products, purity and supervision differ from research-grade vials.
| Who was studied | Dose given | How often | For how long | What happened | |
|---|---|---|---|---|---|
| healthy postmenopausal women (phase 1, single ascending dose) n = 48 |
single dose of 0.02 to 3 mg/kg subcutaneous | once (single dose) | single dose, followed to day 29 and beyond | at 3 mg/kg, total lean mass +3.3% and thigh muscle volume +5.1% at day 29; half-life 10-15 days; injection-site redness | PubMed |
Animal studies: Mouse and marmoset studies gave ACE-031 by injection for 4 to 14 weeks; most abstracts in our pool do not state the mg/kg dose (a rat doping-detection study used 10 mg/kg).
Never approved; development stopped. A placebo-controlled trial in boys with Duchenne muscular dystrophy (PMID 27462804) gave it subcutaneously every 2-4 weeks but was halted for nosebleeds and dilated skin vessels; its abstract does not state the mg/kg dose.
What the research says
ACE-031 (ramatercept) is a soluble activin receptor IIB decoy that blocks myostatin and related ligands. A phase 1 trial in healthy postmenopausal women and a placebo-controlled trial in boys with Duchenne muscular dystrophy both showed it increases lean mass, but the Duchenne trial was stopped early for nosebleeds and dilated skin vessels, and development was discontinued. No trial has shown it improves function.

Myostatin inhibitor ACE-031 treatment of ambulatory boys with Duchenne muscular dystrophy: Results of a randomized, placebo-controlled clinical trial.Humanssize not reportedRandomizedPlacebo-controlledDouble-blindRandomized, double-blind, placebo-controlled, ascending-dose trial; ACE-031 given subcutaneously every 2-4 weeks
In a randomized, double-blind, placebo-controlled ascending-dose trial in ambulatory boys with Duchenne muscular dystrophy, ACE-031 showed non-significant trends toward preserved 6-minute walk distance, higher lean mass and bone density and lower fat mass, but the study was stopped after the second dosing regimen because of epistaxis and telangiectasias.
Key resultNon-significant trend for maintained 6MWT distance in ACE-031 groups vs a decline with placebo; trends toward higher lean mass and BMD and lower fat mass

Blockade of ActRIIB signaling triggers muscle fatigability and metabolic myopathy.Animalssize not reported4 months4-month pharmacological blockade of ActRIIB signaling with soluble ActRIIB-Fc
Four months of soluble ActRIIB-Fc in mice caused extreme muscle fatigability, raised serum lactate, reduced muscle capillarization and a severe metabolic myopathy in the mdx model; the authors caution against this strategy as a sole treatment.
Study details
- Studied
- Mice including the mdx model of Duchenne muscular dystrophy
- Main outcome
- Muscle fatigability, serum lactate, muscle energy metabolism
- Authors' conclusion
- ActRIIB blockade triggered extreme muscle fatigability and a severe metabolic myopathy in mdx mice; the authors caution against deploying this strategy, at least in isolation, for neuromuscular disorders.
- Limitations
- Mouse study; uses soluble ActRIIB-Fc rather than ACE-031 itself.

The cheapest ACE-031 from an independently tested vendor is $8.50 per mg (Andy Peps Studio, AE1 ACE-031 1mg 10 vials, $85.00). Across 11 listings from 10 vendors the median is $20.50 per mg.
Where can I buy ACE-031?
10 vendors tested by Finnrick sell ACE-031. The cheapest per mg right now: Andy Peps Studio ($8.50), Baohua Dongnuo Biotechnology BHD ($8.50), LN Peptides ($8.50). 0 of them publish a lab report (COA) for ACE-031; compare lab scores and reports in the table above before buying.
How much bacteriostatic water for 1 mg of ACE-031?
Adding 1 mL of bacteriostatic water to a 1 mg vial of ACE-031 gives 1 mg/mL: each 10 units on a U-100 insulin syringe holds 100 mcg. With 1 mL it's 1 mg/mL (100 mcg per 10 units); with 3 mL, 0.333 mg/mL (33.3 mcg per 10 units). More water makes small amounts easier to measure. Mixing charts: 1 mg.
Is ACE-031 FDA approved?
Development discontinued. Research use only.
Can ACE-031 be compounded in the US?
Not an FDA-approved drug and not nominated for FDA's 503A bulks list, so US pharmacies can't legally compound it. Compounding status for every peptide.
Does ACE-031 work? What does the research say?
