3 ppm pal-KTTKS in a moisturizer topical (face) | not stated in abstract | 12 weeks | significant reduction in wrinkles/fine lines vs the same moisturizer without the peptide | PubMed |
Animal studies: In a rodent wound-healing study, patches containing 0.1 mg or 1 mg Matrixyl and a cream containing 1 mg were applied for 21 days.
Used as a cosmetic ingredient, not a drug. A 21-woman crow's-feet trial (PMID 36909866) applied a palmitoyl pentapeptide-4 cream twice daily for 8 weeks but its abstract does not give the concentration.
What the research says
Matrixyl (palmitoyl pentapeptide-4, pal-KTTKS) has two small human trials: a 93-woman split-face RCT found less wrinkling than with placebo moisturizer, and a 21-woman trial was too small to be conclusive. The rest of the evidence is cell-culture and animal work showing collagen stimulation.

- Main outcome
- Wound area, collagen density, angiogenesis, re-epithelialization
- Sample
- Animals randomly divided into seven groups; total number not stated
- Authors' conclusion
- Matrixyl cream and patch improved wound healing, with the 1 mg doses exceeding the commercial dressing, and the patch gave higher re-epithelialization than the cream.
- Limitations
- Animal model; species and group sizes not stated in the abstract.

Effect of Palmitoyl-Pentapeptide (Pal-KTTKS) on Wound Contractile Process in Relation with Connective Tissue Growth Factor and α-Smooth Muscle Actin Expression.Cells / labn = 3 for the quantified experimentIn vitro study of Pal-KTTKS (0-0.5 uM) on fibroblast-to-myofibroblast transformation
In fibroblast cultures, pal-KTTKS at 0.1 µM halved myofibroblast transformation (alpha-SMA-positive cells 75% vs 38.6%), but 0.5 µM had no significant effect; authors caution about balancing healing and pro-fibrotic effects.
Key resultalpha-SMA-positive stress fibers fell twofold with 0.1 uM Pal-KTTKS (75 +/- 7.1 vs 38.6 +/- 16.1%, n = 3, p < 0.05); not significant at 0.5 uM
Study details
- Studied
- Cultured fibroblasts and 3D collagen lattices
- Compared with
- Untreated fibroblasts
- Main outcome
- alpha-SMA-positive cells, CTGF, lattice contraction
- Authors' conclusion
- Pal-KTTKS at 0.1 uM reduced alpha-SMA expression and myofibroblast transformation in a dose-dependent manner; a balance between wound-healing and pro-fibrotic properties should be considered.
- Limitations
- In vitro with n = 3; effect lost at higher concentration.

Study details
- Studied
- 3T3-NIH fibroblasts
- Compared with
- Free pentapeptide; 1 mM ascorbic acid
- Main outcome
- Collagen and extracellular matrix synthesis
- Authors' conclusion
- Liposomal pal-KTTKS stimulated collagen production more than free pentapeptide and ascorbic acid, and liposomes could improve peptide stability and skin delivery.

- Compared with
- Vaseline gauze; cellulose dressing
- Main outcome
- Wound closure, inflammation, collagen deposition, angiogenesis
- Authors' conclusion
- Pal-KTTKS-enriched composites improved burn wound healing, best when combined with mesenchymal stem cells.
- Limitations
- Rat study; combined dressing/stem-cell product.

Matrixyl: questions people ask
What is Matrixyl?
Cosmetic collagen peptide. Palmitoyl pentapeptide-4, a signal peptide used in creams to stimulate collagen production.
How much does Matrixyl cost?
The cheapest Matrixyl from an independently tested vendor is $0.104 per mg (Ion Peptide, Palmitoyl Pentapeptide-4 (Raw) – 1000mg, $104.25). Across 10 listings from 9 vendors the median is $0.450 per mg.
Where can I buy Matrixyl?
9 vendors tested by Finnrick sell Matrixyl. The cheapest per mg right now: Ion Peptide ($0.104), Atomik Labz ($0.175), Andy Peps Studio ($0.450). 0 of them publish a lab report (COA) for Matrixyl; compare lab scores and reports in the table above before buying.
How much bacteriostatic water for 10 mg of Matrixyl?
Adding 2 mL of bacteriostatic water to a 10 mg vial of Matrixyl gives 5 mg/mL: each 10 units on a U-100 insulin syringe holds 500 mcg. With 1 mL it's 10 mg/mL (1,000 mcg per 10 units); with 3 mL, 3.333 mg/mL (333.3 mcg per 10 units). More water makes small amounts easier to measure. Mixing charts: 10 mg.
Is Matrixyl FDA approved?
Cosmetic ingredient.
Does Matrixyl work? What does the research say?
Evidence grade B (some human evidence), from 9 papers: One 93-woman split-face RCT found fewer wrinkles and a 21-woman trial was inconclusive; the rest is cell and animal work. See the papers.
What dose of Matrixyl was used in studies?
Matrixyl is studied in people. Caucasian women aged 35-55 with photoaged facial skin (split-face, placebo-controlled): 3 ppm pal-KTTKS in a moisturizer not stated in abstract. These are amounts used in published studies or official labels, not a recommendation.
